Seeds of rice (Oryza sativa L.) cultivar Khao Dawk Mali 105 were cultured on LS (Linsmaier and Skoog, 1965) medium supplemented with 2 mg/l 2,4-D and 0.5 mg/l kinetin. Callus was initiated from these seeds within 7 days. The cultured seeds were harvested at various times, fixed, sectioned, stained and examined microscopically. The histological origin of the callus revealed that callus formation occurred from cells with dense cytoplasm in vascular tissue of mesocotyl and epithelium tissue of scutellum.
