Saccharomyces cerevisiae TISTR 5088 cultivated at room temperature in the shaker (200 rpm) for 36 hours in YEPD medium had 9.0 g/l cell dry weight. After cell homogenization and extraction the yield from the cell wall was 13.65% of cell dry weight. The beta-glucan was extracted from the cell wall by 2 methods : (1) extraction with 50 mM phosphate buffer (pH 8.0 at 90oC) provided crude beta-glucan 20.0 % of cell wall with total sugar 129.0 mg/g and (2) extraction with 0.75 M sodium hydroxide at 75oC provided crude beta-glucan 30.0% of cell wall with total sugar 82.2 mg/g. Puri-fication of the 0.02 g/ml beta-glucan solution by Sephadex G-100 column (1.0 x 110 cm) contained 2.85 mg/ml total sugar. Separation by thin layer chromatography showed that samples had the same Rf as the standard beta-glucan.
Four test diets were formulated, 8.4 g of yeast cell wall /1 kg feed, crude beta-glucan 1.70 g/1 kg feed, 1.0 g of pure beta-glucan/1 kg feed and control feed. The shrimp fed with pure beta-glucan diet showed the highest number of total haemocyte count of 9.901.52 x 104 cell/ml and exhibited better clearance ability after injection with Vibrio harveyi as well as higher disease resistance against V. harveyi (90.0%) when compare to shrimp fed with other test diet.
