Saccharomyces cerevisiae were used as the foreign particles for tissue and cell examination and monitoring of the cellular defense system in black tiger shrimp (Penaeus monodon Fabricius) using the total hemocyte count (THC) and histological techniques. From the experiment, the THC rapidly decreased within 1 hour post injection, from 3.361.85 x 104 cell/mm3 in control to 2.121.96 x 104 cell/mm3 in treatment trial. Histological study showed that the hemocytes and fixed phagocyte in the gill tissue, lymphoid organ, heart tissue, hepatopancreas and connective tissue were sensitive to the foreign particles, but could be detected very slightly in muscle and nervous tissue. The mecha-nism of defense started with yeast particles being attacked by a single hemocyte or fixed phagocyte, followed by an increasing number of cells aggregated to form a nodule and capsule respectively, and finally undergoing melanization and removal from the body. The fine structure and the differential of hemocytes and fixed phagocytes are discussed in this paper.
