Chitosan-collagen hydrogels as controlled release drug carriers

Chitosan hydrogels containing triamcinolone accetonide (TA) were formulated with collagen as controlled release matrices. The hydrogels of 1 : 2 chitosan-collagen containing 5-20% TA were prepared. The release behaviour of the drug from the freeze-dried chitosan-collagen matrices followed Higuchi's mechanism and was pH-dependent. The fast drug release was found at pH 1.2 while the slower drug release at pH 7.4 and 9.0 conformed the Higuchi's model over 5 days (k = 4.03 h-1/2, r2 = 0.984 at pH 7.4 and k = 9.98 h-1/2, r2 = 0.984 at pH 9.0). With increasing the drug loading from 5-20% the release of TA was more prolonged.
