Alkaline lipase production by thermophilic bacteria form the hot-spring in Yala province

Two soil samples and six water samples collected from hot spring in Yala province. Sixty- three isolates of thermophilic bacteria from these samples were found to produce lipase on alkaline medium. Isolate YS02A2 and YS02A3 showed a high lipase activity. Two isolates were gram nega-tive and rod shape. Isolate YS02A2 has white colony while isolate YS02A3 show yellow colony on agar plate containing 2.0% (v/v) olive oil as a carbon source. The experiment were performed in medium containing 2.0 (v/v) coconut oil, 1.0% (w/v) peptone and 1.0% (w/v) yeast extract at 65oC and pH 9.0 on shaker speed 60 rpm. It was found that the maximum activities of alkaline lipase from isolate YS02A2 and YS02A3 could be increased to 34.7 and 38.2 units/ml., respectively at the 60th hr of cultivation.
