In vitro culture of the banana Musa (AAA group, 'Gros Michel') 'Kluai Hom Thong' shoot tip

The use of shoot tip culture for banana micropropagation is described. The Musa (AAA group, 'Gros Michel') 'Kluai Hom Thong' suckers were the source of explants. Culturing small excised shoot apices on modified MS solid medium supplemented with various concentrations of cytokinins induced multiple shoots. Different types of cytokinin, such as N6-benzyladenine (BA), thidiazuron (TDZ) and coconut water (CW) were assessed for their ability to support shoot multiplication. Approximately 4.5 axillary shoots formed at the test concentrations of 4.4 mM BA combined with 15% CW. BA was superior to TDZ for better shoot induction. Auxin was not necessary for root formation. Rooted shoots were hardened and successfully established in soil.
