Rapid and simple sex determination by PCR using SRY and ATL1 primers

We describe a PCR technique for sex determination using 2 pairs of primers, the SRY locus on the Y chromosome and the ATL1 locus on the X chromosome to serve as an internal control of amplification. The PCR reaction yields 2 fragments of 254 bp SRY and 300 bp ATL1 derived fragments from the Y and X chromosome DNA, respectively. We have tested on 24 peripheral blood DNA samples and 6 amniotic fluid cells DNA which were requested for cytogenetic analysis. The amplification results totally corresponded with the cytogenetic analysis. Therefore, we concluded that the described PCR method here is rapid, simple and reliable for the genotypic sex determination.
